The Bruker Microflex is a matrix-assisted laser
desorption/ionization time-of-flight
mass spectrometer (MALDI-TOF MS) primarily
used for the rapid identification of microorganisms,
biomolecules (such as proteins and peptides),
and polymers. It belongs to Bruker's MALDI Biotyper series.
Key components:
Laser source: nitrogen or solid-state laser (often 337 nm UV),
Ionization source: vacuum chamber with MALDI sample holder,
Flight tube: measures the flight time of the ions,
Detector: converts the arrival times of the ions
into a mass spectrum, MALDI sample holder:
metal plate for applying the matrix-mixed samples.
Operation (simplified): Sample preparation:
The analyte is mixed with a MALDI matrix
(e.g., a-cyano-4-hydroxycinnamic acid).
Laser ionization: The laser strikes the matrix
and ionizes the sample.
Ion acceleration: Ions are accelerated into the TOF analyzer.
Detection: The time of flight is measured;
smaller ions reach the detector faster.
Spectrum: Mass/charge ratio (m/z) is plotted against intensity.
